Clomiphene citrate rejuvenates delta cells of anterior pituitary gland following distortion by kolaviron in female wistar rats. (#385)

This research work accessed the roles of clomiphene citrate on the alterations posed on delta cells of anterior pituitary gland by kolaviron (Kv). Garcinia kola has anti-inflammatory property which altered ovulation while clomiphene citrate is useful in the maturation and rupture of the ovary. Thirty adult female Wistar rats were used for this study; divided into six groups A to F with five animals each. While group A received placebo as control, groups B and C received 200 and 100 mg/kg body weight Kv orally once and twice daily for 14 days respectively; group D received 0.50 mg/kg clomiphene citrate orally for 5 days; groups E and F received 200 and 100 mg/kg Kv for 14 days followed by 0.50 mg/kg clomiphene citrate for 5 days respectively. Animals were anaesthesized with chloroform inhalation and 5 ml blood were taken by cardiac puncture to assay for Luteinizing Hormone (LH), Follicle Stimulating Hormone (FSH), Estradiol and Progesterone while the brain removed weighed and fixed in 10% formal calcium immediately for demonstration of H&E and special stains using Nissl for hypothalamus and Aldehyde Fuschin Thionin for delta cells of anterior pituitary gland. The results showed that the delta cells of the anterior pituitary gland were distorted but recovered with administration of clomiphene citrate. The results showed that kv significantly (P<0.05) reduced serum concentrations of FSH, LH and progesterone but significantly (P>0.05) increased the serum concentrations of estradiol. It was concluded that altered delta cells of anterior pituitary gland as a result of kolaviron administered to female wistar rats were reversed with clomiphene citrate which could be implicated in fertility status.
